Study on Improve Diluting Fluid for Concurrent Direct Counts of Bosinophils and Total Leukocytes in Canize Blood



Abstract
The study was conducted in an attempt to estimate the efficiency of various diluting fluids for eosinophil count to oxalated blood and EDTA blood of human and canine, and to prepare the, improved diluting fluid adaptable to concurrent direct counts of eosinophils and total leucocytes of canine blood.
The results, obtained in this experiment were summarized as follows:
1. Hinkelman¢¥s solution and Pilot¢¥s solution were adaptable only for a direct count of eosinophils, and Randolph¢¥s solution and Lee¢¥s solution were adaptable for the concurrent direct count of eosinophils and total leukocytes. Lee¢¥s solution among above four solutions was the best either to direct eosinophil count and to total leukocyte count. However, these solutions wore better to human blood than to canine blood, and ¢¥especially better to EDTA blood than to collated blood in both of hu ian and canine.
2. Among these four diluting fluids, Lee¢¥s solution. was recommended to be improved for can current direct count of eosinophils and total leukocytes of canine blood. The formula of improved solution obtained, through ¢¥ the experiment was as follows. In this fluid, eosinophils were stained y reddish brown in the part of eosinophilic gronules and. blue in other oart of the cells, basophils were stained dark blue like as a lump of black granules and other leukocytes, were stained blue, showing each cells better distinctly¢¥ than in Lee¢¥s solution.
-Propylene glycol 50 ml
Solution I ? Distilled Water 40 ml
-1% Phloxine Aqueous Solution 10 ml
Solution k 1% Methylene blue in Methyl Alcohol 5 ml
Mix 100 ml of Solution I and 5m1 of Solution 11 thoroughly and filter before us
